ABSTRACT

Objective@#To evaluate the effect of semaphorin 3A (Sema3A) pre-treated bone marrow mesenchymal stem cells (BMSC) sheets on new bone formation in type 2 diabetes mellitus rats.@*Methods@#Type 2 diabetes mellitus (T2DM) were induced by injection of streptozotocin, and the BMSC were isolated, controlled, identified and induced into cell sheets. Fifteen T2DM rats were randomly divided into control, sheets and Sema3A-sheets group and the calvarial critical size defect (CSD) model of rats were established. The defect zone of rats from control group were implanted with bone powder. The defect zone of rats from sheets group were implanted with bone powder and BMSC sheets. The defect zone of rats from Sema3A-sheets group were implanted with bone powder and BMSC sheets pretreated with 1.0 mg/L Sema3A. After 8 weeks, the bone samples were harvested and analyzed by micro-CT scanning, HE staining for the evaluation of new bone formation, and the immunohistochemical analysis for the expression of osteogenesis-related proteins including type Ⅰ collagen (COL- Ⅰ ), bone morphogenetic protein-2 (BMP-2), and osteocalcin (OCN).@*Results@#The BMSC were isolated and cultured, and oil red O and Alizarin red S staining proved the multi-potential differentiation. Eight weeks after the establishment of calvarial CSD model, Sema3A-sheet group showed the most abundant new bone formation (0.516±0.070), with increased bone volume fraction, namely bone volume/tissue volume (BV/TV) compared with sheets group (0.319±0.050) and control group (0.224±0.037) (P<0.05), and the sheets group showed increased BV/TV compared with control group (P<0.05). While trabecular thickness (Tb.Th) control group showed no difference in three groups (P>0.05). HE staining also confirmed that Sema3A-sheets group showed the most new bone formation. Sheet group (0.174±0.051) compared showed difference with control group (0.099±0.033) (P< 0.05), and Sema3A-sheet group (0.421±0.069) showed increased bone formation compared with sheet group and control group (P<0.05). Immunohistochemistry showed that BMSC sheet increased the expression of osteogenesis-related proteins including COL-Ⅰ, BMP-2 and OCN, while Sema3A pretreatment showed more obvious increase of the expression of COL-Ⅰ and OCN.@*Conclusions@#The combined implantation of bone powder and Sema3A stimulated BMSC sheets significantly increased bone regeneration in vivo. Therefore, Sema3A pre-treated BMSC sheets transplantation provides a new strategy for restoring bone defect in T2DM.

ABSTRACT

The implant prosthesis has been extensively used in clinic recently, and implant failure is appearing. Many factors may cause the failure, and they work together generally. This paper summarizes and analyzes the failure cases related to implant treatment and relevant risk factors of oral implants in Department of Implantation, School of Stomatology, The Fourth Military Medical University during the past six years, in order to improve the success rate of implant prosthesis and provide guidance for clinical application.

ABSTRACT

Objective:To explore the calvarial critical size defect (CSD)in rats with type 2 diabetes mellitus(T2DM).Methods:T2DM model of SD rats(weighted 300-320 g)was induced by high fat and high sugar diet and low dose intraperitoneal streptozotocin (STZ)injection.The rats with T2DMand the normal controls were divided into 4 groups(n=3)respectively.Defects with the diame-ter(mm)of 2,3,4 and 5 were made on the central calvaria of each rat.General observation,X-ray examination and histological study were performed 8 weeks postoperatively.Results:In the T2DM group,only the defects of 2 mm diameter were healed completely,X-ray resistance and new bone formation were observed;the defects of 3,4 and 5 mm diameter were unhealed,X-ray transmission was observed and newly formed bone was insufficient.In the control group,the defects of 2,3 and 4 mm diameter were healed completely, X-ray resistance and new bone formation were observed;the defects of 5 mm diameter were unhealed,X-ray transmission was ob-served,newly formed bone was insufficient.Conclusion:The calvarial CSD of T2DM rat model can be defined as the defect with the diameter of 3 mm.

ABSTRACT

Objective:To evaluate the outcome of implant-supported all-ceramic fixed partial prostheses (FPPs)in posterior area. Methods:The clinical data of 1 20 implants-supported 53 fixed dentures in 47 patients were collected from July 201 1 to June 201 2 and prospectively studied.Complication and failure of implants and /or prosthesis,biological and technical complications were evaluated. Results:43 restorations with 1 00 implants in 37 cases were followed up for 1 2 -24 months.Veneering ceramic chipping was observed in 9(20.9%)prostheses.Inflamed marginal gingivitis was found around 3(7.0%)prostheses.No implant was involved in technical complication.Cumulative survival rate was 1 00% for implant-based analysis and 1 00% for prostheses-based analysis.Conclusion:Implant-supported all-ceramic fixed partial dentures may be a feasible treatment modality for posterior dental restoration.

ABSTRACT

<p><b>OBJECTIVE</b>To evaluate the clinical and radiographic outcomes of placing dental implants in the posterior maxilla using the transalveolar technique.</p><p><b>METHODS</b>Between January 2005 and December 2009, a total of 94 cases, 55 males and 39 females, aged (46.9 ± 11.8) years, were consecutively enrolled in this study with maxillary sinus floor augmentation using the transalveolar technique. Radiographic evaluations were conducted on panoramic and periapical radiographs at each recall. The follow-up clinical examination included cumulative survival rate(CSR) of implants, peri-implant marginal bone loss (MBL) and the height of sinus floor augmentation and the endo-sinus bone gain (ESBG).</p><p><b>RESULTS</b>Sinus membrane perforation was found in 4 cases.Finally 126 implants (90 cases) were placed. The perforation rate of this surgical procedure was 3.08% (4/130).Four implants lost during the healing time, the early success of implants was 96.80% (121/125). During a mean follow-up time of (34.4 ± 20.4) months, all the implants were successfully in function, with the cumulative survival rate after function loading of 100.00%. The mean MBL was (0.75 ± 0.51) mm during the healing time, and (0.48 ± 0.41) mm during the follow-up period. The mean residual bone height (RBH) was (7.27 ± 1.30) mm at the position of implants placement, the length of implant protruding into the sinus was (2.77 ± 1.15) mm, and the height of sinus floor augmentation was (4.52 ± 1.39) mm. At the loading time, the mean endo-sinus bone gain was (3.81 ± 1.60) mm. After the follow-up time of 34 months, the newly formed bone on the maxillary sinus floor underwent further remodeling with a median bone reduction value of 0.37 (0.10, 0.88) mm.</p><p><b>CONCLUSIONS</b>Maxillary sinus floor augmentation using the transalveolar technique is a predictable treatment modality. The augmentation of the maxillary sinus floor using the transalveolar technique could lead to bone formation under the sinus. The newly formed bone showed slight absorbtion in the long term follow-up.</p>

Resonance frequency analysis(RFA) has been successfully applied to measure the stability of implants, and proved to an objective method. Domestic and international scholars in the field of organ implantation have paid great attention on RFA in recent years. The present paper systematically introduces principles and applications of RFA in buccal medicine. The advantages and insufficiencies of RFA are discussed too.

ABSTRACT

BACKGROUND: Human periodontal ligament fibroblasts (HPLF) is the crucial cells in maintaining the configuration and function of periodontium. Adverse stress may cause HPLF to synthesize more inflammatory agents, which may cause the damage of periodontium.OBJECTIVE: To investigate the role of p38 MAPK of HPLF in the expres sion of inflammatory cytokine of interleukin-6 (IL-6) subjected to mechanical pressure, and explore the mechanism of the occlusal trauma to periodontium.DESIGN: A randomized and controlled trial.SETTING: Pathological Laboratory of the Fourth Military Medical Univer sity of Chinese PLA.MATERIALS: The HPLF were obtained from the middle part of 1/3 pe riodontium of 12 to 16-year-old youth whose 20 healthy permanent premo lar teeth should be extracted for orthodontic need. Main reagents and ap paratus: IL-6 enzyme-linked immunoabsorbent assay (ELISA) kit (Staff Room of Immunology of the Fourth Military Medical University of Chinese PLA); ELISA apparatus (Huadong Electronic Tube Factory); p38 MAPK specific inhibitor of SB203580 (produced by Biochemical Company, ob tained as a present from Professor Jiang, Staff Room of Pathophysiology, Southern Medical University).METHODS: The cells were primarily cultured till the 4-5 passages, and randomly divided into four groups: ①pressure-loading control group: the cell s were not subjected to pressure-loading and without pretreatment; ② pressure-loading group: the cells were subjected to continuous pressure-load ing (200 kPa) but without pretreatment; ③ pretreatment control group: the supernatant were added with 10 g/L dimathyl sulfoxide (DMSO, SB203580 solvent) at 1 hour before pressure-loading, the method and time of pressure loading were the same as those in the pressure-loading group; ④ pretreated group: the cells were pretreated with 1 μmol/L SB203580 (a specific in hibitor of the p38 MAPK) at 1 hour before pressure-loading, the method and time of pressure-loading were the same as those in the pressure-loading group. The cytosol and the supernatant in each group were sampled at 16 and 24 hours after pressure-loading respectively. The IL-6 expressions at different time points were detected with ELISA.MAIN OUTCOME MEASURES: The amount of IL-6 expression in HPLF induced by pressure with or without pretreatment by SB203580, a specific inhibiter of p38 MAPK.RESULTS: The expressions of IL-6 after continuous pressure-loading for 16 and 24 hours in the pressure-loading group were (143.1±0.42) and (49.46±1.01) ng/L, which were obviously higher than those in the pres sure-loading control group [(18.36±0.43), (18.78±0.50) ng/L, P < 0.05]. The expressions of IL-6 after continuous pressure-loading for 16 and 24 hours in the pretreatment group were (56.39±0.72) and (21.52±1.39) ng/L, which were obviously higher than those in the pressure-loading control group [(137.96±0.54), (48.47±0.79) ng/L, P < 0.05].CONCLUSION: p38 MAPK of HPLF acts as important cooperative mechanism to regulate IL-6 synthesis induced by mechanical pressure.

ABSTRACT

<p><b>OBJECTIVE</b>To investigate the effect of human serum albumin (HSA) on cell attachment of human gingival epithelial cells (HGE).</p><p><b>METHODS</b>HGE were primary cultured with keratinocyte serum-free medium (KSFM) and dispase. The cultured cells were immunohistochemically stained by monoclonal anti-pan cytokeratin. MTT test was employed to investigate the influence of HSA on the cell attachment on polystyrene surface. The cell growth curve of HGE which were cultured in KSFM with 50 g/L HSA was observed.</p><p><b>RESULTS</b>The results showed significant decrease in cell numbers within 8 hours after HGE were inoculated, in which the polystyrene surface was preincubated with 50 g/L HSA. But it did not prove to be the case from 10 hours to 24 hours after HGE were inoculated. There were no significant difference within 24 hours in cell numbers between cultured in KSFM with 50 g/L HSA and control. The cell numbers in cell growth curve of HGE in KSFM with and without 50 g/L HSA did not show significant difference.</p><p><b>CONCLUSIONS</b>HSA preincubation on polystyrene were produce inhibitory effect of HGE attachment in early stage.</p>

ABSTRACT

<p><b>OBJECTIVE</b>The objective of this study was to investigate the tarnish induced by Streptococcus sobrinus serotype g on Co-Cr, Ni-Cr and casting titanium alloys.</p><p><b>METHODS</b>Co-Cr, Ni-Co and casting titanium alloys were respectively machined into a size of 10 mm x 10 mm x 1 mm in tablets, and totally 90 specimens with 30 of each were prepared for the test. The 30 specimens of each alloy were randomly and equally divided into three subgroups, including the blank control group, the media control group and the inoculated media group. Under the aerobic condition, the surfaces of the sucrose agar media were inoculated with Streptococcus sobrinus serotype g. The labeled sterile specimens were placed on the surface of the media for 10 weeks. The specimens were changed to a new sucrose media with bacteria each week. The specimens were removed after 10 weeks of incubation, immersed in 0.05% glutaraldehyde solution to eliminate AaY4, rinsed with distilled water. After 2 weeks, the specimens were removed and observed visually or examined by MINOLTA CR-100 color apparatus.</p><p><b>RESULTS</b>Compared with the blank control, the control media without bacteria did not influence the alloy specimens (P > 0.05). Streptococcus sobrinus serotype g caused tarnish on specimens. There was significant difference between the control media group and the blank control group with inoculated media group on all the specimens (P < 0.05).</p><p><b>CONCLUSION</b>Streptococcus sobrinus serotype g affects the color of Co-Cr, Ni-Cr and casting titanium alloys, but the color does not change from yellow-green range to red-yellow range.</p>

ABSTRACT

objective: To investigate the effect of human serum albumin(HSA)on the attachment of human gingival epithelial cells(HGEs)to commercial pure titanium(cpTi).Methods: HGEs were cultured, the cells of passage 2~5 were seeded on to cpTi samples which were preincubated in keratinocyte serum free medium with 50 mg/ml HSA or without HSA. Cell attachment was studied by immunofluorescence analysis.Results: HGEs were cultured and identified by positive expression of cytokeratin. 4,12 and 24 h after seeding attached cells on HSA preincubated cpTi were 218,730 and 849, those on the control 417,745 and 864, respectively.Conclusion: The early stage of cell attachment of HGEs on to cpTi may be inhibited by HSA.

ABSTRACT

Objective: To obtaine the parameters of tooth mobil ity in people with normal health teeth . Methods:Tooth mob ility of 1 4, 1 4 in 12 subjects (7 males and 5 females, aged 21 to 23 years old) was measured by FX-1 tooth mobility measurement system 6 times a day for 5 days.The data were statisticaly analyzed. Results: The average tooth mobility parameters Q and R varied significantl y (P0.05).Conclusion: The tooth mo bility parameters of S,K,t1 and t2 can steadily reflect the changing of tooth mobility.

ABSTRACT

Objective: To study the potential of growth and mineralization of cultured osteoblasts on the indium tin oxide conductive(ITO) glass disk coated with polypyrrole film (Ppy) . Methods: Osteoblasts were seeded onto ITO glass or ITO glass coated with polypyrrole film of implant materials. The cells were stained by HE and Von Kossa, observed by light microscope and scanning electron microscope (SEM) on day 3, 7, 14 and 28 after seeding respectively. Results: The osteoblast cells attached well to both ITO surfaces and Ppy films, mineralized nodules was observed on Ppy film. Conclusion: The polypyrrole films have good bone biocompatibility and it can be used as the biomaterials for dental implant.

?Objective: To study the difference in the mobility of health teeth between Chinese young male and female. Methods: Tooth mobility (TM) of all teeth at right side was measured with FX 1 TM measuring system in 100 young males and 100 young females of Han nationality.Slope (K) value was calculated from the measurements. Results: TM values of the teeth in the females were significantly heigher than those in males ( P 0.05) Conclusion: Difference in tooth mobility exists between healthy young male and female of Chinese Han nationality.

ABSTRACT

Objective:Through the cases retrospective study, to analyze the correlation between the adjacent endodontic teeth and the retrograde implantitis. Methods: 32 patients were selected and there were 39 implants (ITI 28,MDIC 11), which adjacent teeth had received endodontic therapy. Record the time from endodontic therapy and the distance of the apex of implants to the adjacent tooth by X-ray photograph. Results: There were 10 implants with retrograde peri-implantitis of 39 implants. In 8 implants, their adjacent teeth received root canal therapy within 3 months(P
